General description
Mucin Degradation Enzyme is a metalloprotease useful for digesting heavily O-glycosylated mucin proteins. This product is made for preparative scale digestions of viscous material that is dense with mucin-type glycoproteins.
Application
For use as a sample preparation tool in research and discovery. Used for proteolytic degradation of mucin-dense samples to allow for downstream processing.
Biochem/physiol Actions
The Mucin Degradation is based on a bacterial protease that specifically recognizes densely glycosylated mucins and mucin-domains, and proteolytically cleaves the peptide backbone. In nature this presumably enhances the virulence of the pathogenic microbe, but in the laboratory is a useful way of accessing molecular structures in tissues normally shielded by mucins.

- Tool for viscosity reduction and tissue breakdown of mucin containing samples.
- Not inhibited by most general protease inhibitors such as AEBSF, Aprotinin, Bestatin, E-64, Pepstatin and Leupeptine. When needed digests can be done in presence of the ReadyShield® Protease Inhibitor Cocktail PIC0002.
- Mucin Degradation Enzyme can be used to liberate soluble mucin fragments from cells or tissue without mechanical disruption or reduction.
- Mucin Degradation Enzyme can be used for enzymatic hydrolysis of mucins in the isolation of glyco-RNA samples.
Physical form
Product is supplied as lyophilized powder containing the indicated amount of protein.
Preparation Note
Solution of Mucin Degradation Enzyme can be prepared by reconstitution of the lyophilized powder at a concentration of 1-5mg/ml protein. See CoA for lot specific percent protein in powder. The reconstituted protein solution may be clear or hazy and is stable for 2 weeks at 2-8 °C. For longer storage, aliquot the protein solution and store at -20 °C. The enzyme solution should be protected from direct light.
Storage and Stability
The product retains activity for at least 2 years when stored lyophilized at -20°C.
Other Notes
For applications that require more precision with highly purified enzyme like glycoproteomic experiments please consider using Mucinase StcE (SAE0202) and Mucinase SmE (SAE0220).
