Biochem/physiol Actions

PDLIM4 (PDZ and LIM domain 4) is an actin associated protein consisting of a LIM (Lin11/Isl1/Mec3) domain and PDZ (PSD-95/Dlg/ZO-1) domain. It is generally expressed within the cytokine cluster. It is a tumor suppressor gene, which actively suppresses Src activation through interacting with Src and PTPL1 (Protein tyrosine phosphatase 1). As a result, it leads to a series of Src inactivation cycle. Therefore, silencing of the gene directly disrupts the Src inactivation cycle followed by aberrant Src activation in human cancers. It also consists of two putative transcription start points (tsp) responsible for potential binding of transcriptional factors. Mutation in the PDLIM4 is associated with radial bone mineral density (BMD) and osteoporosis.
