Quality Control Testing
Evaluated by Western Blotting in lysate from HeLa cells treated with UV.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected phospho-Histone H2A (Ser129) in lysate from HeLa cells treated with UV.
Tested Applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected p-Histone H2A in Yeast Extract.
Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected p-Histone H2A in HeLa cells serum-starved overnight -/+ Etoposide 20ug/mL for 3 hours.
Peptide Inhibition Assay: Target band detection in UV treated HeLa cells was prevented by preblocking of a representative lot with the immunogen phosphopeptide, but not the corresponding non-phosphopeptide.
A 1:1,000 dilution from a representative lot detected Histone H2A in HeLa treated with UV.

Histone H2A.1 (UniProt: P04911; also known as H2A) is encoded by the HTA1 (also known as H2A1, SPT11, YD9934.10, YDR225W) gene (Gene ID: 851811) in Saccharomyces cerevisiae (yeast). Histone H2A is one of the 5 main histones that are involved in the structure of chromatin in eukaryotic cells. Featuring a main globular domain and a long N terminal tail, H2A is involved with the structure of the nucleosomes of the ′beads on a string′ structure. Histones are modified post-translationally by acetylation, phosphorylation, methylation, and ubiquitination and these modifications regulate DNA transcription, repair, recombination, and replication. Histone H2A.1 undergoes phosphorylation at Serine 129 that plays a crucial role in various cellular processes involved in DNA repair, genomic stability, and cell cycle regulation. Yeast histone H2A1 contains a [ST]-Q motif (aa 129-130) in the C-terminal region that is phosphorylated by the Mec1 and Tel1 kinases in response to DNA double strand breaks or DNA damage signals. It serves as a recognition mark for the recruitment of DNA repair factors and facilitates the repair of damaged DNA. Overall, Histone H2A.1 phosphorylation at serine 129 is a significant event in maintaining genomic integrity and cell homeostasis. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our
reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
