Spécificité

Clone 2F12 is a ZooMAb® rabbit recombinant monoclonal antibody that detects Lysine-specific demethylase 2B (KDM2B/FBXL10). It targets an epitope within 19 amino acids from the internal part of the protein.

Immunogène

KLH-conjugated linear peptide corresponding to 19 amino acids from the internal part of human KDM2B/FBXL10.

Application

Quality Control Testing

Evaluated by Western Blotting in C6 nuclear extract.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected KDM2B/FBXL10 in C6 nuclear extract.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected KDM2B/FBXL10 in HeLa Nuclear Extract.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected KDM2B/FBXL10 in human cerebral cortex and skin tissue sections.

Affinity Binding Assay Analysis: The binding affinity of this antibody to KDM2B/FBXL10 peptide was at least one thousand-fold (1,000X) higher than that obtained with the non-specific control peptide.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected KDM2B/FBXL10 in A-431, HeLa and HepG2 cells.

Description de la cible

Lysine-specific demethylase 2B (UniProt: Q8NHM5; also known as EC:1.14.11.27, JmjC domain-containing histone demethylation protein 1B) is encoded by the KDM2B (also known as CXXC2, Fbl10, FBXL10, JHDM1B, PCCX2) gene (Gene ID: 84678) in human. Lysine-specific demethylase 2B (KDM2B/FBXL10) is a protein that plays a role in the regulation of gene expression through its demethylase activity. KDM2B/FBXL10 is a member of the Jumonji C (JmjC) domain-containing demethylase family. It consists of several functional domains, including JmjC domain responsible for histone demethylation, CXXC-type zinc finger domains that bind to DNA, and a PHD finger domain involved in protein-protein interactions. KDM2B/FBXL10 functions as a histone demethylase, targeting specific lysine residues on histone proteins. It is involved in regulating key biological processes such as cell cycle progression, development, and differentiation. Its expression is dynamically regulated during development, suggesting its importance in embryogenesis and tissue homeostasis. Aberrant expression or mutations in KDM2B gene have been associated with different types of cancer, including breast, lung, and prostate cancer. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Forme physique

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.

Stockage et stabilité

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
