Application

Quality Control Testing

Evaluated by Western Blotting in lysate from L6 cells treated with insulin.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Phosphoepitope SR proteins in lysates from L6 cells treated with insulin (100 nM; 5 min) but not in untreated cells.

Tested Applications

Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected Phosphoepitope SR proteins in HeLa cells.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Description de la cible

Ser-Arg-rich (SR) proteins make up a family of functionally and structurally conserved phosphoproteins that are required for alternative and constitutive pre-mRNA splicing. These proteins are involved in various aspects of pre-mRNA splicing and in spliceosome assembly including the identification and appropriation of splice-sites, and the manipulation of alternative splicing regulation. During mitosis, SR proteins are diffusely distributed in the cytosol, but later in mitosis, nuclear speckles are assembled into cytoplasmic structures known as mitotic interchromatin granules that disappear around telophase, concomitant with the reconstruction of nuclear speckles just after nuclear envelope/lamina formation. These proteins are characterized by their modular composition consisting of two domains of interest: an N-terminal RNA recognition motif (RRM) that serves in the determination of DNA-binding specificity, and the RS domain, an arginine and serine rich, C-terminal domain that serves as a shuttling and localization director of SR proteins and as a splicing activation domain. The RS domains are extensively phosphorylated, and this phosphorylation is essential for nuclear import of SR proteins and for their functions in mediating spliceosome assembly. Partial dephosphorylation of SR proteins is also critical for progression of the assembled spliceosome to catalysis and for a series of post-splicing events from the interaction with the mRNA transport machinery to SR protein-mediated translational control. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Forme physique

Purified recombinant mouse monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.

Stockage et stabilité

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
