Spécificité

Clone 4B5 is a mouse monoclonal antibody that detects Tau. It targets an epitope within 27 amino acids from the C-terminal half. It preferentially reacts with 3R tau isoforms, however, it also displays slight reactivity with 4R tau isoforms.

Immunogène

KLH-conjugated linear peptide corresponding to 27 amino acids from the C-terminal half of human Tau.

Application

Quality Control Testing

Evaluated by Western Blotting with recombinant, human Tau, isoform 2N3R.

Western Blotting Analysis: A 1:500 dilution of this antibody detected recombinant, human Tau protein, isoform 2N3R.

Tested Applications

Western Blotting Analysis: A representative lot detected Tau (MTBR3R) in Western Blotting application (Kanaan, N.M., et al. (2020). Nat Commun. 11(1):2809).

ELISA Analysis: A representative lot detected Tau (MTBR3R) in ELISA application (Kanaan, N.M., et al. (2020). Nat Commun. 11(1):2809).

ELISA Analysis: A representative lot detected Tau recombinant protein, isoform 2N4R (blue line) and 2N3R (red line) (Data courtesy of Nicholas Kanaan, Ph.D., Michigan State University, USA).

Immunohistochemistry Applications: A representative lot detected Tau (MTBR3R) in Immunohistochemistry applications in human temporal cortex tissues from an Alzheimer s disease patient (Data courtesy of Nicholas Kanaan, Ph.D., Michigan State University, USA).

Description de la cible

Microtubule-associated protein tau (UniProt: P10636; also known as Neurofibrillary tangle protein Paired helical filament-tau (PHF-tau)) is encoded by the MAPT (also known as MAPTL, MTBT1, TAU) gene (Gene ID: 4137) in human. Tau is a highly abundant protein found predominantly in the nervous system. Its primary role is to stabilize and support microtubule assembly, which are essential for maintaining the structure and function of nerve cells. It is present throughout the brain, particularly in regions involved in memory and cognition. MAPT gene consists of multiple exons that undergo alternative splicing, resulting in the generation of different isoforms that differ in the number and arrangement of repeats in the microtubule-binding domain. The expression of the MAPT transcripts varies and depends on the stage of neuronal maturation and neuron type. Aberrant aggregation and accumulation of tau protein have been linked to several neurodegenerative diseases collectively known as tauopathies. The most common tauopathy is Alzheimer′s disease, where hyperphosphorylated tau protein forms neurofibrillary tangles inside neurons. Other tauopathies include frontotemporal dementia, progressive supranuclear palsy, and cortico-basal degeneration. These diseases are characterized by cognitive decline, motor disturbances, and memory loss.
