General description

ZooMAb® antibodies represent an entirely new generation of recombinant monoclonal antibodies. Specificity

Clone 1B21 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects PD-L2 (CD273). It targets an epitope within 25 amino acids from the extracellular domain within the N-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 25 amino acids from the extracellular domain within the N-terminal half of human PD-L2 (CD273).

Application

Quality Control Testing

Evaluated by Western Blotting in Jurkat cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected PD-L2/CD273 in Jurkat cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected PD-L2/CD273 in EL4, and Raji cell lysates, and 1:10,000 dilution with His-tagged recombinant fragment of Human PD-L2/CD273 with a C-terminal IgG Fc linker.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected PD-L2/CD273 in Jurkat cells.

Flow Cytometry Analysis: 1 µg from a representative lot detected PD-L2/CD273 in one million Jurkat cells.

Affinity Binding Assay: A representative lot of this antibody bound PD-L2/CD273 peptide with a KD of 5.7 x 10-8 in an affinity binding assay.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Programmed cell death 1 ligand 2 (UniProt: Q9BQ51; also known as PD-1 ligand 2, PD-L2, PDCD1 ligand 2, Programmed death ligand 2, Butyrophilin B7-DC, B7-DC, CD273) is encoded by the PDCD1LG2 (also known as B7DC, CD273, PDCD1L2, PDL2) gene (Gene ID: 80380) in human. Three isoforms of PD-L2 have been reported that are produced by alternative splicing. Isoforms 1 and 2 are single-pass type I membrane proteins and isoform 3 is a secreted protein. PD-1 and PD-1 ligands 1 and 2 (PD-L1 and PD-L2) are B7:CD28 family members that regulate T cell activation and peripheral tolerance. When engaged together with the TCR, the interaction of PD-1 with its ligands delivers an inhibitory signal to T cell proliferation and cytokine production. While PD-L1 is broadly expressed in hematopoietic and non-hematopoietic cells, PD-L2 expression is highly restricted to antigen presenting cells (APCs), including dendritic cells (DCs) and macrophages. The PD-1 pathway plays a key role in the progressive loss of effector T cell responses during chronic HIV infection. Under some conditions, blockade of this pathway can restore many T cell functions. PD-L2 is initially produced with signal peptide (aa 1-19) that is removed to produce the mature protein that contains a large extracellular region (aa. 20-220), a transmembrane domain (aa 221-241), and a cytoplasmic tail (aa 242-273). This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
