Specificity

Clone 2J18 is a ZooMAb® recombinant rabbit monoclonal antibody that specifically detects ZIP-14. It targets an epitope within 21 amino acids from the N-terminal, extracellular domain.

Immunogen

KLH-conjugated linear peptide corresponding to 21 amino acids from the N-terminal, extracellular domain of human ZIP14.

Application

Quality Control Testing

Evaluated by Western Blotting in rat liver tissue lysate.

Western Blotting Analysis (WB): A 1:1,000 dilution from a representative lot detected ZIP14 in lysate from rat liver tissue.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected ZIP14 in lysate from human heart tissue.

Affinity Binding Assay: A representative lot of this antibody bound with ZIP14 peptide with at least one thousand-fold (10,000X) higher affinity than with non-specific control peptide.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected ZIP14 in human kidney tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected ZIP14 in HepG2 cells.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Metal cation symporter ZIP14 (UniProt: Q15043; also known as Metal cation symporter ZIP14, LIV-1 subfamily of ZIP zinc transporter 4, LZT-Hs4, Solute carrier family 39 member 14, Zrt- and Irt-like protein 14, ZIP-14) is encoded by the SLC39A14 (also known as KIAA0062, ZIP14) gene (Gene ID: 23516) in human. The ZIP14 gene is mapped to human chromosome 8p21.3. The encoded protein is a transmembrane metal ion transporter and is a member of the LIV-1 subfamily. ZIP-14 is a multi-pass membrane protein and contains six transmembrane domains (TMDs), a metalloprotease, and a histidine-rich motif. It is predominantly expressed in the liver, pancreas, fetal liver, thyroid gland, heart and weakly expressed in spleen and thymus. ZIP-14 primarily functions as a divalent metal transporter specific for zinc (Zn). However, some studies report, it also mediates the transport of manganese (Mn), iron (Fe), and cadmium (Cd). As a zinc importing protein, ZIP-14 has an important role in glucose homeostasis. ZIP-14 along with ZIP8 are prime Mn2+ importers of the brain s capillary bed and in human brain microvascular endothelial cells (HBMVEC) in in-vitro conditions. Mutations in the ZIP14 gene lead to manganese dyshomeostasis, which in turn results in manganese accumulation in childhood-onset parkinsonism dystonia patients. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
