Specificity

Clone 2C9 is a ZooMAb^® rabbit recombinant monoclonal antibody that detects CDC20. It targets an epitope within 22 amino acids from the internal region.

Immunogen

KLH-conjugated linear peptide corresponding to 22 amino acids from the internal region of human CDC20.

Application

Quality Control Testing

Evaluated by Western Blotting in human testis tissue lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected CDC20 in human testis tissue lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected CDC20 in human tonsil tissue lysate.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected CDC20 in human tonsil tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected CDC20 in A-431 cells.

Affinity Binding Assay: A representative lot of this antibody bound with CDC20 peptide with at least one million-fold (1,000,000X) higher than with non-specific control peptide.

Target description

Cell division cycle protein 20 homolog (UniProt: Q12834; also known as Cdc20, p55CDC) is encoded by the CDC20 gene (Gene ID: 991) in human. CDC20 contains an N-terminal region with a C-box, KEN-box, Mad2 interacting motif (MIM), and CRY-box motif, and a C-terminal region with seven WD40 repeats and an Ile-Arg (IR) motif. CDC20 plays a critical role regulating cell cycle progression and apoptosis. It acts as an activator of the anaphase-promoting complex (APC) and forms the APCCdc20 E3 ubiquitin ligase complex that is responsible for tagging specific proteins for degradation, thus facilitating mitosis. The APCCdc20 ubiquitin signaling pathway involved in other biological processes as well, including dendritic morphogenesis, brain development and plasticity in the mammalian brain. Cdc20 overexpression is associated with tumorigenesis and poor prognosis. Its downregulation is associated with DNA damage, impaired repair process, and apoptosis. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
