Specificity

Clone 2D23 is a ZooMAb^® rabbit recombinant monoclonal antibody that specifically detects CD5L. It targets an epitope within 25 amino acids from the N-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 25 amino acids from the N-terminal half of human CD5L.

Application

Quality Control Testing

Evaluated by Western Blotting with His/ABP-tagged recombinant fragment of Human CD5L.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected His/ABP-tagged recombinant fragment of Human CD5L.

Tested Applications

Enzyme Immunoassay (ELISA) Analysis: A serial of dilutions from a representative lot detected His/ABP-tagged recombinant fragment of Human CD5L.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected CD5L in Human spleen tissue sections.

Affinity Binding Assay:: A representative lot of this antibody bound His/ABP-tagged recombinant fragment of Human CD5L with a KD of 2.7 x 10-8 in an affinity binding assay.

Target description

CD5 antigen-like (UniProt: O43866; also known as Apoptosis inhibitor expressed by macrophages, hAIM, CT-2, IgM-associated peptide, SP-alpha) is encoded by the CD5L (also known as AP16, Q203/PRO229) gene (Gene ID: 922) in human. CD5L is a secreted glycoprotein of the SRCR superfamily that is synthesized with a signal peptide (aa 1-19), which is subsequently cleaved off to produce the mature form. Its expression is transcriptionally controlled by LXRs and its expression is observed in macrophages, spleen, lymph node, thymus, bone marrow, and fetal tissue. However, it is not detected in non-lymphoid tissues. It circulates in serum in relatively high amounts and serves as a serum biomarker for inflammatory conditions. It is involved in a broad spectrum of biological functions. It participates in the pathogenesis of inflammatory processes by preventing the apoptosis of macrophages and other cell types. It also acts as a key regulator of lipid synthesis. CD5L is reported to reduce lipid droplet size in adipocytes. Following incorporation into mature adipocytes via CD36-mediated endocytosis, it associates with cytosolic fatty acid synthase activity and inhibits its activity and leads to lipolysis, degradation of triacylglycerols into glycerol and free fatty acids. It participates in obesity-associated inflammation following recruitment of inflammatory macrophages into adipose tissues, a cause of insulin resistance and obesity-related metabolic disorders. This ZooMAb^® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
