Specificity

Clone 1E12 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Transcriptional activator Myb (MYB). It targets an epitope within 17 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 17 amino acids from the C-terminal region of human Transcriptional activator Myb (MYB).

Application

Quality Control Testing

Evaluated by Western Blotting in MOLT-4 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected MYB in MOLT-4 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected MYB in Jurkat and RAW264.7 cell lysates.

Affinity Binding Assay: A representative lot of this antibody bound c-Myb peptide with a KD of 8.4 x 10-7 in an affinity binding assay.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected MYB in human tonsil tissue sections.

Target description

Transcriptional activator Myb (UniProt: P10242; also known as Proto-oncogene c-Myb) is encoded by the MYB gene (Gene ID: 4602) in human. MYB is a proto-oncogene transcription factor that specifically recognize the sequence 5′-YAAC[GT]G-3′. It functions as a master regulator of gene expression in diverse cell types, including hematopoietic cells where it controls cell proliferation and differentiation. It exhibits a tripartite structure consisting of three domains: an N-terminal DNA-binding domain, a centrally located transcriptional activation domain, and a C-terminal domain that is involved in transcriptional repression. MYB is negatively regulated by microRNA-155. It can undergo phosphorylation on multiple sites that induce its proteasomal degradation. Its phosphorylation by homeodomain interacting protein kinase 1 (HIPK1) reduces its transcription factory activity but not the protein level. Aberrant expression of MYB gene is reported in acute myeloid leukemia. The leukemic functions of MYB are reported to be mediated by CBP/P300 co-activation of a distinct set of transcription factor complexes. These MYB complexes assemble aberrantly with LYL1, E2A, C/EBP family members, LIM domain only 2 (LMO2), and special AT-rich sequence-binding protein-1 (SATB1). This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
