General description
NUAK family kinase 1(NUAK1)/AMPK-related protein kinase-5 (ARK5) belongs to the human AMP-activated protein kinases (AMPKs) family. It is highly expressed in the heart and brain, and at lower levels in skeletal muscle, kidney, ovary, placenta, lung, and liver.
Biochem/physiol Actions
NUAK family kinase 1(NUAK1)/AMPK-related protein kinase-5 (ARK5) facilitates cancer cell migration activity. It plays a major role in tumorigenesis, cancer invasion, and metastasis. This tumor cell survival factor is activated by AKT. It serves as an ATM kinase under conditions of nutrient starvation. Overexpression of ARK5 is observed in human ovarian cancer. It is also overexpressed in breast cancer, colorectal carcinoma, and hepatocellular carcinoma (HCC). ARK5 knockdown prevents the aggressive features of SKOV3 cells. NUAK1 is involved in tolerance to glucose starvation and suppresses Fas-induced apoptosis by phosphorylation of caspase 6 (CASP6), thus suppressing the activation of the caspase and the subsequent cleavage of CFLAR.

We recently demonstrated that the LKB1 tumour suppressor kinase, in complex with the pseudokinase STRAD and the scaffolding protein MO25, phosphorylates and activates AMP-activated protein kinase (AMPK). A total of 12 human kinases (NUAK1, NUAK2, BRSK1, BRSK2, QIK, QSK, SIK
