General description
Glucose activates sphingosine kinase 2 in pancreatic β cells leading to a rise in S1P levels, which is important for glucose-stimulated insulin secretion. Human sphingosine kinase 2 (GenBank Accession No.NM_020126), is full length with an N-terminal His tag, has a molecular weight of 74 kDa, and is expressed in a Baculovirus infected Sf9 cell expression system.
Application
Sphingosine kinase 2 is useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling. Sphingosine kinase 2 has been used in a study to assess neuroapoptosis in the developing mouse brain. Sphingosine kinase 2 assays were used to confirm FTY720 bioactivity in a study investigating cranial allograft incorporation.
Unit Definition
One unit of sphingosine kinase activity is defined as the amount of enzyme required to produce 1 pmol of S1P/min
Physical form
Formulated in 25 mM Tris-HCl, pH 8.0, 100 mM NaCl, 0.05% Tween-20, 50% glycerol and 3 mM DTT.
Preparation Note
Thaw on ice. Upon first thaw, briefly spin tube containing enzyme to recover full content of the tube. Aliquot enzyme into single use aliquots. Store remaining undiluted enzyme in aliquots at -70°C. Note: Enzyme is very sensitive to freeze/thaw cycles.

Endogenous stem cell recruitment to the site of skeletal injury is key to enhanced osseous remodeling and neovascularization. To this end, this study utilized a novel bone allograft coating of poly(lactic-co-glycolic acid) (PLAGA) to sustain the release of FTY720, a

Recent studies suggest that sphingolipid metabolism is altered during type 2 diabetes. Increased levels of the sphingolipid ceramide are associated with insulin resistance. However, a role for sphingolipids in pancreatic beta cell function, or insulin production, and release remains to

Our previous studies have indicated that de novo ceramide synthesis plays a critical role in ethanol-induced apoptotic neurodegeneration in the 7-day-old mouse brain. In this study, we examined whether the formation of sphingosine 1-phosphate (S1P), a ceramide metabolite, is associated
