General description

Cell cycle checkpoint protein RAD17 is encoded by the gene hRAD17, in humans. hRAD17 is a 69kDa protein, which is made of 590 amino acids. RAD17 was first identified in Schizosaccharomyces pombe, and its homolog in humans is hRAD17. Human RAD17 gene is localized to the chromosomal region 5q13.

Application

Biochem/physiol Actions

RAD17 is a DNA damage cell cycle checkpoint protein. RAD17 forms a complex with four small replication factor c (RFC) subunits namely, Rfc2, Rfc3, Rfc4 and Rfc5. This complex, called RFC-related complex, acts as a clamp loader and detects DNA damage. Once DNA damage is detected, RFC-related complex recruits 9-1-1 complex (Rad9, Rad1 and Hus1). This, in turn, activates ATM, ATR, Chk1 and Chk2 kinases. Therefore, RAD17 plays an essential role in maintaining genomic stability. RAD17 might be a potential tumor suppressor gene and may have a role in the progression of head and neck squamous cell carcinoma (HNSCC) by contributing to genomic instability.
