Specificity

Clone 701 is a mouse monoclonal antibody that specifically detects SCUBE1 but not, SCUBE2 or SCUBE3. It targets an epitope within 200 amino acids from the C-terminal region.

GST-tagged, recombinant fragment from the C-terminus of human SCUBE1.

Application

Quality Control Testing

Evaluated by Western Blotting in HEK-293T cells transfected with pFlag-hSCUBE1.

Target description

Signal peptide, CUB and EGF-like domain-containing protein 1 (UniProt: Q8IWY4) is encoded by the SCUBE1 gene (Gene ID: 80274) in human. The gene encodes a platelet-endothelial cell surface protein that is expressed during early embryogenesis. The protein plays a role during the early stages of central nervous system development. SCUBE1 is a novel adhesive molecule that mediates platelet matrix interaction and ristocetin-induced platelet agglutination. It is produced with a signal peptide sequence at the N-terminus (aa 1-23) followed by nine tandemly arranged epidermal growth factor (EGF)-like repeats (aa 33-402), a spacer region and three cysteine-rich repeat motifs and a CUB domain at the C-terminus (aa 798-910). SCUBE1 protein is highly expressed in platelets and stored in the platelet -granules, and translocated to the surface upon platelet stimulation. Studies suggest that matrix-bound and soluble fragments of SCUBE1 may play critical (patho)physiological roles in cardiovascular biology. Platelet activation plays a crucial role in acute coronary syndrome (ACS) and acute ischemic stroke (AIS), and platelet stimulation is associated with increased plasma concentration of SCUBE1. Studies show plasma SCUBE1 concentration is significantly elevated in ACS and AIS but, not in coronary artery disease (CAD) patients. Plasma SCUBE1 is implicated to be a potential biomarker of platelet activation in acute thrombotic disease. It may also function in renal regeneration and repair. (Ref.: Tu, C-F., et al. (2006).
