Target description

Tumor necrosis factor receptor superfamily member 13C (UniProt: Q96RJ3; also known as CD268, B-cell-activating factor receptor, BAFF receptor (BAFF-R),BLyS receptor 3) is encoded by the TNFRSF13C (also known as BAFFR, BR3) gene (Gene ID: 115650) in human. BAFF-R/CD268, is a single pass, type III membrane protein in the tumor necrosis factor receptor (TNFR) superfamily. It features an extracellular domain (1-78) with three cysteine-rich motifs, a transmembrane domain (79-99), and a short cytoplasmic tail (100-184). CD268 is crucial for B-cell survival, differentiation, and proliferation. It mediates signaling pathways, including NF- B and MAPK, promoting cell survival and antibody production upon ligand binding. BAFF has been shown to have profound effect on T cell costimulation, through BAFF-R/CD268 as well. CD268 is implicated in diseases like multiple myeloma, B-cell lymphomas and autoimmune diseases. Its overexpression supports malignant plasma cell survival, making it a target for therapeutic interventions in these conditions. (Ref.: Ng, L.G., et al. (2004). J Immunol. 173(2):807-17.)
