Application

Quality Control Testing

Evaluated by Western Blotting in lysate from Jurkat cells treated with PMA and Ionomycin.

Western Blotting Analysis (WB): A 1:500 Dilution of this antibody detected phospho-CARD11 (Ser893) in lysates from Jurkat cells treated with Phorbol 12-myristate 13-acetate (PMA; 15 ng/mL) and Ionomycin (1 g/mL) for 20 minutes.

Tested Applications

Western Blotting Analysis: A 1:500 dilution from a representative lot detected phospho-CARD11 (Ser893) in lysates from Jurkat cells treated with Phorbol 12-myristate 13-acetate (PMA; 15 ng/mL) and Ionomycin (1 g/mL) for 20 minutes.

Immunoprecipitation Analysis: A representative lot detected phospho-CARD11 (Ser893) in Immunoprecipitation applications (Kutzner, K., et al. (2022). Sci Signal. 15(723):eabk3083).

Western Blotting Analysis: A representative lot detected phospho-CARD11 (Ser893) in Western Blotting applications (Kutzner, K., et al. (2022). Sci Signal. 15(723):eabk3083).

Target description

Caspase recruitment domain-containing protein 11 (UniProt: Q9BXL7; also known as CARD-containing MAGUK protein 1, Carma 1) is encoded by the CARD11 (also known as CARMA1) gene (Gene ID: 84433) in human. CARD11 belongs to the membrane-associated guanylate kinase (MAGUK) family, characterized by the presence of a caspase activation and recruitment domain (CARD) (aa 18-110), a coiled-coil domain (aa 130-449), a membrane-binding domain (PDZ) (aa 667-755) and a guanylate kinase-like domain (aa 973-1140). These structural features enable CARD11 to function as a scaffold protein, facilitating the assembly of signaling complexes in immune cells. The protein undergoes post-translational modifications, including phosphorylation, that promotes the assembly of the CARD11-BCL10-MALT1 (CBM) complex and lymphocyte activation. Phosphorylation can also suppress CARD11 functions in T or B cells. There are several constitutive and inducible CARD11 phosphorylation sites in T cells. Phosphorylation of Ser893 of CARD11 prevents the activation of the transcription factor NF- B, the kinase JNK, and the protease MALT1. Mutations of the CARD11 gene or dysregulation of CBM complex signaling causes combined immune deficiency and can lead to uncontrolled cell proliferation and survival, contributing to tumorigenesis. Additionally, CARD11 is associated with autoimmune diseases due to its critical role in regulating immune responses. (Ref: Kutzner, K., et al. (2022). Sci Signal. 15(723):eabk3083)
