General description

Specificity

Clone 2B9 is a ZooMAb® rabbit recombinant monoclonal antibody that detects Smac/DIABLO. It targets an epitope within 184 amino acids from human Smac/DIABLO.

Application

Quality Control Testing

Evaluated by Western Blotting in A549 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Smac/DIABLO in A549 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected Smac/DIABLO in K-562 cell lysate.

Immunocytochemistry Analysis: A 1:1,000 Dilution from a representative lot detected Smac/DIABLO in A549 cells.

Flow Cytometry Analysis: 1 µg from a representative lot detected Smac/DIABLO in one million Jurkat cells.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Diablo IAP-binding mitochondrial protein (UniProt: Q9NR28; also known as Diablo IAP-binding mitochondrial protein, Diablo homolog, mitochondrial, Direct IAP-binding protein with low pI, Second mitochondria-derived activator of caspases, SMAC, Diablo IAP-binding mitochondrial protein, cleaved form) is encoded by the DIABLO (also known as SMAC) gene DIABLO (Gene ID: 56616) in human. Smac/DIABLO, is a mitochondrial protein that is released into the cytosol in response to apoptotic signals. It has a distinct structure with an N-terminal mitochondrial targeting sequence that is cleaved upon mitochondrial release, exposing the IAP-binding motif. The primary function of Smac/DIABLO is to promote apoptosis by antagonizing inhibitor of apoptosis proteins (IAPs). IAPs bind to and inhibit caspases, which are essential for the execution of apoptosis. This activity is crucial for regulating cell death and maintaining cellular homeostasis. Smac/DIABLO is ubiquitously expressed in various tissues. Expression levels can be influenced by cellular stress and apoptotic signals. In cancer, the ability of tumor cells to evade apoptosis is a hallmark of disease progression, and alterations in Smac/DIABLO expression or function can influence this process. Therapeutically, Smac/DIABLO mimetics are being explored as potential cancer treatments to enhance apoptosis in tumor cells. Additionally, dysregulation of Smac/DIABLO has implications in neurodegenerative diseases and other conditions where apoptosis plays a key role. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
