Specificity

Clone 2E4 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Transmembrane protein 106B (TMEM106B). It targets an epitope within 22 amino acids from the N-terminal, cytoplasmic domain.

Immunogen

KLH-conjugated linear peptide corresponding to 22 amino acids from the N-terminal, cytoplasmic domain of human Transmembrane protein 106B (TMEM106B).

Application

Quality Control Testing

Evaluated by Western Blotting in Jurkat cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected TMEM106B in Jurkat cell lysate.

Tested Applications

Affinity Binding Assay: A representative lot of this antibody bound TMEM106B peptide with a KD of 1.1 x 10-7 in an affinity binding assay.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected TMEM106B in Human testis and Rat brain tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected TMEM106B in U2OS cells.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Transmembrane protein 106B (UniProt: Q9NUM4; also known as TMEM106B) is encoded by the TMEM106B gene (Gene ID: 54664) in human. TMEM106B is a single-pass type II membrane protein that contains a cytoplasmic domain (aa 2-96), a transmembrane domain (aa 97-117), and a lumenal domain (aa 118-274). Its expression is observed in the brain, including the frontal cortex. Some expression has also been reported in lung epithelial cells. In neurons, it is involved in the transport of late endosomes/lysosomes and may also be involved in dendrite morphogenesis and maintenance by regulating lysosomal trafficking. It may serve as a molecular brake for retrograde transport of late endosomes/lysosomes, possibly via its interaction with MAP6. It also plays a role in the regulation of lysosomal size and responsiveness to stress and is required for lysosomal acidification. A truncated C-terminal fragment of TMEM106B (aa 120-354) has also been reported that aggregates into stable amyloid fibrils in the brain of subjects suffering from diverse genetic and sporadic tauopathies, amyloid-β amyloidoses, and synucleinopathies. It also plays a role in ACE2-independent SARS-CoV-2 entry into cell where it is required for post-endocytic stage of virus entry by facilitating spike-mediated membrane fusion. Mutations in TMEM106B gene have been linked to Ubiquitin-positive frontotemporal dementia (UP-FTD), an autosomal dominant neurodegenerative disease. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
