Specificity

Clone 2K17 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects APPL1. It targets an epitope within 19 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 19 amino acids from the C-terminal region of human APPL1.

Application

Quality Control Testing

Evaluated by Western Blotting in HeLa cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected APPL1 in HeLa cell lysate.

Tested Applications

Affinity Binding Assay: A representative lot of this antibody bound APPL1 peptide with a KD of 1.4 x 10-6 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected APPL1 in HeLa cells.
Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected APPL1 in human tonsil tissue sections.

Target description

DCC-interacting protein 13-alpha (UniProt: Q9UKG1; also known as Dip13-alpha, Adapter protein containing PH domain, PTB domain and leucine zipper motif 1) is encoded by the APPL1 (also known as APPL, , DIP13A, KIAA1428) gene (Gene ID: 26060) in human. APPL1 is a multifunctional homodimeric adapter protein that binds to various membrane receptors, nuclear factors, and signaling proteins to regulate many processes, such as cell proliferation, immune response, endosomal trafficking, and cell metabolism. Its expression is observed in heart, ovary, pancreas, and skeletal muscle. It is reported to regulate signaling pathway leading to cell proliferation through interaction with RAB5A and subunits of the NuRD/MeCP1 complex. APPL1 is overexpressed in many cancers and has been implicated in tumorigenesis and tumor progression. It is shown to promote the migration of gastric cancer cells by regulating Akt2 phosphorylation. In cells with siRNA silenced APPL1, cell migration is inhibited. APPL1 also functions as a positive regulator of innate immune response via activation of AKT1 signaling pathway by forming a complex with phosphoinositide-3-kinase regulatory subunit 1 (PIK3R1). It is also involved in trafficking of the TGF-β receptor type 1 from the endosomes to the nucleus via microtubules in a TRAF6-dependent manner. It is also involved in regulating adiponectin and insulin signaling pathways. APPL1 is phosphorylated at serine 410 by protein kinase A and this phosphorylation severely impairs its binding to Inositol polyphosphate-5-phosphatase (OCRL). Mutations in APPL1 gene are linked to maturity-onset diabetes of the young 14 (MODY14), an autosomal dominant inheritance that onsets in childhood  or early adult hood. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
