Specificity

Clone 1G14 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects B-cell lymphoma/leukemia 11B (Bcl-11B). It targets an epitope within 16 amino acids from the internal region.

Immunogen

KLH-conjugated liner peptide corresponding to 16 amino acids from the internal region of human B-cell lymphoma/leukemia 11B (Bcl-11B).

Application

Quality Control Testing

Evaluated by Western Blotting in Jurkat cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected Bcl-11B in Jurkat cell lysate.

Tested applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected Bcl-11B in MOLT4 cell lysate.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Bcl-11B in Jurkat cells.

Affinity Binding Assay: A representative lot of this antibody bound Bcl-11B with a KD of 1.3 x 10-7 in an affinity binding assay.

Target description

B-cell lymphoma/leukemia 11B (UniProt: Q9C0K0; also known as BCL-11B, B-cell CLL/lymphoma 11B, COUP-TF-interacting protein 2, Radiation-induced tumor suppressor gene 1 protein, hRit1) is encoded by the BCL11B (also known as CTIP2, RIT1) gene (Gene ID: 64919) in human. Bcl-11B is a zinc finger protein transcription factor plays a critical role in the development, proliferation, differentiation, and survival of T cells. It is highly expressed in brain and in malignant T-cell lines derived from patients with adult T-cell leukemia/lymphoma. Bcl-11B works as both a genetic suppressor and activator, acting directly, attaching to promoter regions, as well as indirectly, attaching to promoter-bound transcription factors. It contains six C2H2-type zinc finger domains. Of those, domains 3 and 4 bind to the DNA, while other regions are important for protein interactions. It is shown to be a key regulator of both differentiation and survival of T-lymphocytes during thymocyte development in mammals. It also plays a role in controlling the responsiveness of hematopoietic stem cells to chemotactic signals by modulating the expression of CCR7 and CCR9, which direct the movement of progenitor cells from the bone marrow to the thymus. Bcl-11B also acts as a tumor suppressor that represses transcription through direct, TFCOUP2-independent binding to a GC-rich response element. In a murine model. The loss of a BCL11B gene increases susceptibility to thymic lymphoma in murine models and to acute lymphoblastic leukemia (T-ALL) in human. Bcl-11B also plays a role in fetal neuronal development, with important roles for the differentiation and development of various neuronal subtypes in the central nervous system. This ZooMAbZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
