General description

Matrin-3 (UniProt P43243) is encoded by the MATR3 (also known as ALS21, KIAA0723, VCPDM) gene (Gene ID 9782) in human. Matrin-3 is an inner nuclear matrix protein involved in multiple nuclear processes, including DNA double strand break response, DNA replication/repair, chromatin organization, and RNA stability and processing. Matrin-3 contains two RNA recognition motifs (a.a. 398-473/RRM1 and a.a. 496-571/RRM2) that mediate RNA-binding and a DNA-binding zinc finger motif (a.a. 801-832). Matrin-3 is known to interact with the RNA-binding protein TDP-43 and MATR3 gene mutations are linked to amyotrophic lateral sclerosis (ALS). Matrin-3 is also a known target of multiple alpha herpes viruses and is known to be utilized by HIV-1 as a Rev cofactor.

Specificity

Expected to react with both spliced isoforms of human MATR3 reported by UniProt (P43243).

Immunogen

GST-tagged recombinant internal fragment from the N-terminal half of human martin-3.

Application

Immunohistochemistry Analysis: An 1:250 dilution from a representative lot detected MATR3 in human cerebellum, human small intestine, and mouse cerebral cortex tissue sections.
This Anti-MATR3 Antibody, clone 3G1.1 is validated for use in Western Blotting, Immunohistochemistry (Paraffin) for the detection of MATR3.

Quality

Evaluated by Western Blotting in SHSY-5Y cell lysate.

Western Blotting Analysis: 0.5 µg/mL of this antibody detected MATR3 in 10 µg of SHSY-5Y cell lysate.

Target description

~110 kDa observed. 94.62 kDa (isoform 1) and 62.58 kDa (isoform 2) calculated. The detected band size is consistent with the reported apparent molecular weight (Nakayasu, H., and Berezney, R. (1991). Proc. Natl. Acad. Sci. U. S. A. 88(22):10312-10316).
