Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Ganglioside GD2 in human lung cancer tissue sections.

Tested Applications

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Ganglioside GD2 in IMR-32 cells.

Flow Cytometry Analysis: 0.1 µg from a representative lot detected Ganglioside GD2 in one million IMR-32 cells.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Disialoganglioside GD2 (GD2) is a glycosphingolipid, which is part of the ganglioside family. Its structure consists of a ceramide (a fatty acid linked to a sphingosine) backbone connected to a complex oligosaccharide chain that includes two sialic acid residues. GD2 is expressed at low levels in normal tissues, primarily on the surface of neurons and peripheral nerve fibers. However, it is highly expressed in certain types of cancer cells, particularly in neuroblastoma, melanoma, osteosarcoma, and small cell lung cancer. GD2 has complex biological functions in tumor cells, including contributing to cellular adhesion and mediating cytotoxic signals. GD2 plays a crucial role in cell-cell interactions, cell signaling, and modulation of the immune response. It is involved in the regulation of cell growth, differentiation, and adhesion. GD2 is also known to participate in signal transduction pathways by interacting with other cell surface molecules. Tumor cells that are positive for GD2 often have enhanced cell survival, are invasive and proliferative. Furthermore, binding of antibodies leads to immune-independent cell death mechanisms. GD2 contributes to T-cell dysfunction, and functions as an immune checkpoint. Therapeutic strategies, including monoclonal antibodies targeting GD2, have been developed to treat cancers. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant mouse monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
