Quality Control Testing
Evaluated by Western Blotting in NIH/3T3 cell lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected FKHR/FOXO1 in NIH/3T3 cell lysate.
Tested Applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected FKHR/FOXO1 in Jurkat, MCF7, and U-2 OS cell lysates, and FKHR/FOXO1 recombinant protein .
Affinity Binding Assay Analysis: The binding affinity of this antibody to FKHR/FOXO1 peptide was at least ten thousand-fold (10,000X) higher than that obtained with the non-specific control peptide.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected FKHR/FOXO1 in NIH/3T3 cells.

Forkhead box protein O1 (UniProt:Q12778; also known as Forkhead box protein O1A, Forkhead in rhabdomyosarcoma) is encoded by the FOXO1 (also known as FKHR, FOXO1A) gene (Gene ID: 2308) in human. Forkhead box protein O1 ( FKHR/FOXO1) is a transcription factor that belongs to the Forkhead box (Fox) family of proteins. It is characterized by a common Forkhead (or winged helix) domain, a variant of the helix-turn-helix motif. FKHR/FOXO1 protein contains three residues, Thr24, Ser256, and Ser319 that lie within consensus sequences for phosphorylation by protein kinase B (PKB). It plays a critical role in regulating diverse cellular processes, including cell cycle arrest, apoptosis, glucose metabolism, lipid metabolism, oxidative stress responses, and cellular differentiation. It is expressed in numerous organs, including the liver, muscle, adipose tissue, pancreas, and brain. Expression levels and activation of FKHR/FOXO1 can be influenced by various physiological and pathological conditions, such as nutrient availability, hormonal signaling, and cellular stress. Dysregulation of FKHR/FOXO1 activity has been associated with metabolic disorders, including diabetes and obesity. Altered FOXO1 expression or function has also been observed in cancer, neurodegenerative diseases, and cardiovascular diseases. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our
reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
