Diaphanous-related formin 2 (DIAPH2) is part of the diaphanous subfamily of the formin homology family of proteins. It is a nuclear protein. A defect in this gene leads to premature ovarian failure. The gene DIAPH2 is mapped to human chromosome Xq21.33.

생화학적/생리학적 작용

Diaphanous-related formin 2 (DIAPH2) interacts specifically with the distal regulatory element (DRE) of the human skeletal α-actin gene between positions -1260 and -1193, the binding site being an A/T-rich sequence element. The DRF-2 also binds well to the muscle creatine kinase (MCK) muscle-specific regulatory factor, MEF-2 binding site and to the A/T-rich regulatory element of the skeletal muscle fast-twitch troponin C gene. This troponin C site competes for DRF-2 driven expression of the skeletal α-actin gene in C2 cells. The DRF-2 site alone does not activate transcription in muscle cells when linked to the SV40 promoter. DRF-2 binding element is a MEF-2 binding site that is required but insufficient for regulation of muscle-specific skeletal α-actin gene expression by the DRE. Therefore, muscle-specific regulation of the human skeletal α-actin gene requires interactions between the other elements of the composite DRE enhancer with the protein-DNA complex formed by DRF-2.
