Specificity

Clone 2N17 is a ZooMAb® recombinant rabbit monoclonal antibody that detects Maf-B. It targets an epitope within 17 amino acids from the N-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 17 amino acids from the N-terminal half of human Maf-B.

Application

Quality Control Testing

Evaluated by Western Blotting in SK-N-MC cell lysate.


Western Blotting Analysis (WB): A 1:10,000 dilution of this antibody detected Maf-B in SK-N-MC cell lysate.

Tested Applications

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Maf-B in human kidney and human testis tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound Maf-B peptide with at least ten thousand-fold (10,000X) higher than with non-specific control peptide.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Maf-B in SK-N-MC cells.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Transcription factor MafB (UniProt: Q9Y5Q3; also known as V-maf musculoaponeurotic fibrosarcoma oncogene homolog B, Maf-B) is encoded by the MAFB (also known as KRML) gene (Gene ID: 9935) in human. Maf-B is a member of the large Maf family of transcription factors, characterized by a basic leucine zipper (bZIP) domain that facilitates DNA binding and dimerization. This structure enables Maf-B to bind to MARE (Maf recognition elements) sequences within the DNA, allowing it to regulate gene expression. Maf-B can form both homodimers and heterodimers with other bZIP transcription factors, enhancing its regulatory versatility. As a transcription factor, Maf-B plays a critical role in the regulation of gene expression involved in cell differentiation and development. It is particularly important in the differentiation of hematopoietic cells, including monocytes and macrophages, and is also involved in the development of kidney podocytes and islet β cells in the pancreas. Maf-B influences cell cycle regulation, apoptosis, and cellular differentiation processes by activating or repressing target genes. MafB expression is tissue-specific, predominantly found in monocytes, macrophages, and kidney. Its expression is also noted during certain developmental stages, particularly in embryogenesis where it is critical for the proper development of various organs and tissues. In adult tissues, Maf-B expression continues to play a crucial role in maintaining the function and development of specific cell types. In the hematopoietic system, alterations in Maf-B expression are associated with leukemias and other hematological disorders. In the kidney, Maf-B dysfunction can contribute to renal diseases, including glomerulosclerosis. Maf-B′s role in pancreatic β cells also links it to diabetes. Additionally, Maf-B has been implicated in the development of obesity through its regulatory effects on adipogenesis. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
