Specificity

Clone 1G7 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Carnitine palmitoyltransferase 2 (CPT2). It targets an epitope within 18 amino acids from the mitochondrial matrix domain within the N-terminal half

Immunogen

KLH-conjugated linear peptide corresponding to 18 amino acids from the mitochondrial matrix domain within the N-terminal half of human Carnitine palmitoyltransferase 2 (CPT2).

Application

Quality Control Testing

Evaluated by Western Blotting in Rat liver tissue lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected CPT2 in Rat liver tissue lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected CPT2 in lysates from HT-29 cells and Human liver and Rat liver tissues..

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected CPT2 in Human kidney and Human liver tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected CPT2 in HT-29 and MCF-7 cells.

Affinity Binding Assay: A representative lot of this antibody bound CPT2 peptide with a KD of 4.1 x 10-8 in an affinity binding assay.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Carnitine O-palmitoyltransferase 2, mitochondrial (UniProt: P23786; also known as EC:2.3.1.21, Carnitine palmitoyltransferase II, CPT II) is encoded by the CPT2 (also known as CPT1) gene (Gene ID: 1376) in human. CPT2 is a mitochondrial inner membrane protein that is involved in the synthesis of acylcarnitines from accumulated acyl-CoA metabolites. It is synthesized with a transit peptide, which is subsequently cleaved off. It catalyzes the transfer of long- and medium-chain fatty acids from cytoplasm into mitochondria, where oxidation of fatty acids takes place. It reconverts acylcarnitines back into the respective acyl-CoA esters that can then undergo β-oxidation, an essential step for the mitochondrial uptake of long-chain fatty acids and their subsequent β-oxidation. It displays higher activity with medium (C8-C12) and long-chain (C14-C18) acyl-CoA esters. Deficiency of CPT2 can result in the most common inherited disorder of long-chain fatty acid oxidation affecting skeletal muscle. Mutations in CPT2 gene can result in a lethal neonatal form, a severe infantile hepato-cardio-muscular form, or a rather mild myopathic form characterized by exercise-induced myalgia, weakness, and myoglobinuria. In CPT2 deficiency, long‐chain acylcarnitines are transported across the inner mitochondrial membrane but cannot be converted to their corresponding long‐chain acyl‐CoAs. They accumulate in the mitochondrial matrix and are transported out into plasma. These elevated levels may produce cardiac arrhythmias. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonal. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
