Specificity

Clone 1D18 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects S100A1 protein. It targets an epitope within 13 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 13 amino acids from the C-terminal half of human S100A1 protein.

Application

Quality Control Testing

Evaluated by Immunohistochemistry (Paraffin) in Human heart tissue sections.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution of this antibody detected S100A1 in Human heart tissue sections.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected S100A1 in lysate from Human heart tissue.

Immunofluorescence Analysis: A 1:1,000 dilution from a representative lot detected S100A1 in Human heart tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound S100A1 peptide with a KD of 5.7 x 10-6 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected S100A1 in HepG2 cells.

Target description

Protein S100-A1 (UniProt: P23297; also known as S-100 protein alpha chain, S-100 protein subunit alpha, S100 calcium-binding protein A1) is encoded by the S100A1 (also known as S100A) gene (Gene ID: 6271) in human. S100A1 is a small calcium binding protein that plays important roles in several biological processes, including Ca2+ homeostasis, chondrocyte biology, and cardiomyocyte regulation. Typically, it exists as a homodimer, but it can also associate with two β-chains or as one - and one β-chain. It is highly expressed in the heart and serves as an important regulator of the cardiac contractility. In lesser quantities it is also found in skeletal muscle and brain. It is reported to regulate a network in cardiomyocytes controlling sarcoplasmic reticulum Ca2+ cycling and mitochondrial function through interaction with the ryanodine receptors RYR1 and RYR2, sarcoplasmic reticulum Ca2+-ATPase/ATP2A2 and mitochondrial F1-ATPase. It facilitates diastolic Ca2+ dissociation and myofilament mechanics in order to improve relaxation during diastole. S100A1 can also bind zinc in vitro, however, its binding sites are different from the calcium binding sites. Post-translational glutathionylation is shown to increase its affinity for calcium by about 10-fold. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
