Specificity

Clone 3F7 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects BCR-ABL1 phosphorylated on tyrosine 245 of ABL1, isoform 2.

Immunogen

KLH-conjugated linear peptide corresponding to 13 amino acids surrounding phosphotyrosine 245 of human ABL1, isoform 2.

Application

Quality Control Testing

Evaluated by Western Blotting in K562 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected BCR-ABL1 (pTyr245) in K562 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected phospho BCR-ABL1 (Tyr245) in MCF-7 cell lysate.

Affinity Binding Assay: A representative lot of this antibody bound phospho-ABL1 (Tyr245) peptide with a KD of 8.4 x 10-9 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected phospho-ABL1 (Tyr245) in K562 cells.

Peptide Inhibition Assay: Target band detection in a lysate from NIH3T3 cells was prevented by preblocking of a representative lot with the immunogen phosphopeptide, but not the corresponding non-phosphopeptide.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Tyrosine-protein kinase ABL1 (UniProt: also known as EC:2.7.10.2, Abelson murine leukemia viral oncogene homolog 1, Abelson tyrosine-protein kinase 1, Proto-oncogene c-Abl, p150) is encoded by the ABL1 (also known as ABL, JTK7) gene (Gene ID: 25) in human. ABL1 is a non-receptor tyrosine protein kinase that plays a role in many KEY® processes linked to cell growth and survival. It is detected in the nucleus and cytoplasm and shuttles between them depending on environmental signals. It is sequestered into the cytoplasm through interaction with 14-3-3 proteins. In the nucleus it displays DNA-binding activity and is involved in DNA-damage response and apoptosis. It can also localize to mitochondria in response to oxidative stress. ABL1 phosphorylates multiple receptor tyrosine kinases and is reported to promote endocytosis of EGFR, facilitate the formation of neuromuscular synapses through MUSK, and inhibit PDGFRB-mediated chemotaxis. It is also involved in the endocytosis of activated B-cell receptor complexes. ABL1 also regulates multiple pathological signaling cascades during infection. Several known tyrosine-phosphorylated microbial proteins have been identified as ABL1 substrates. ABL1 is stabilized in the inactive form by an association between the SH3 domain and the SH2-TK linker region, interactions of the N-terminal cap, and contributions from an N-terminal myristoyl group and phospholipids. It is activated by autophosphorylation and by SRC-family kinase-mediated phosphorylation. Its phosphorylation at Tyr-70 by members of the SRC family of kinases disrupts SH3 domain-based autoinhibitory interactions and INTERMOLECULAR® associations and enhances its kinase activity. Phosphorylation at Tyr-226 and Tyr-393 also correlate with increased activity. BCR®-ABL1, a constitutively active form of ABL1 is known to cause Chronic myelogenous leukemia (CML) and phosphorylation of Tyr-245 on the ABL1 kinase domain is commonly used as a marker of BCR®-ABL1 activation. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, Affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
