Specificity
Clone 2I3 is a ZooMAb® rabbit recombinant monoclonal antibody that detects HDAC6. It targets an epitope within 21 amino acids from the N-terminal region.
Immunogen
KLH-conjugated linear peptide corresponding to 21 amino acids from the N-terminal region of human HDAC6.
Application
Quality Control Testing
Evaluated by Western Blotting in human testis tissue lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected HDAC6 in human testis tissue lysate.
Tested Applications
Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected HDAC6 in human testis tissue sections.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected HDAC6 in K-562 cells.
Affinity Binding Assay: A representative lot of this antibody bound with HDAC6 peptide at least one thousand-fold (100,000X) higher affinity than with non-specific control peptide.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.
Target description
Histone deacetylase 6 (UniProt: Q9UBN7; EC:3.5.1.98; also known as HD6, Protein deacetylase HDAC6, Tubulin-lysine deacetylase HDAC6) is encoded by the HDAC6 (also known as KIAA0901, JM21) gene (Gene ID: 10013) in human. HDAC6 is a member of the histone deacetylase family, characterized by two catalytic domains that enable its enzymatic activity. It also contains a unique C-terminal region with a ubiquitin-binding domain, which allows it to interact with ubiquitinated proteins, distinguishing it from other HDACs. HDAC6 primarily functions as a deacetylase, removing acetyl groups from lysine residues on histones and non-histone proteins. This activity plays a crucial role in regulating gene expression, protein stability, and cellular signaling pathways. HDAC6 is particularly involved in the deacetylation of tubulin, impacting microtubule dynamics and cellular transport processes. HDAC6 is widely expressed in various tissues, with particularly high levels in the brain, immune cells, and muscle tissues. Its expression can be influenced by cellular stress and inflammatory signals, reflecting its role in stress responses. Alterations in HDAC6 expression and activity have been implicated in several diseases, including cancer, neurodegenerative disorders, and inflammatory conditions. In cancer, HDAC6 can promote tumor progression by deacetylating key proteins involved in cell growth and survival. In neurodegenerative diseases, such as Alzheimer′s, HDAC6 is associated with the clearance of misfolded proteins, suggesting a role in maintaining proteostasis. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Reconstitution
300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
