General description
Dual specificity mitogen-activated protein kinase kinase 7 (EC 2.7.12.2; UniProt O14733; also known as c-Jun N-terminal kinase kinase 2, MAP kinase kinase 7, MAPK/ERK kinase 7, MAPKK 7, MEK 7, JNK-activating kinase 2, JNK kinase 2, JNKK 2, SAPK kinase 4, SAPKK-4, SAPKK4, Stress-activated protein kinase kinase 4) is encoded by the MAP2K7 (also known as JNKK2, MEK7, MKK7, PRKMK7, SKK4) gene (Gene ID 5609) in human. MKK7 belongs to the family of mitogen-activated protein kinase kinases (MAPKKs) involved in mediating cellular signal transduction in response to proinflammatory cytokines stimulation and environmental stresses. MKK7 and the related MKK4 function as crucial upstream transducers in the stress-activated protein kinase/c-Jun N-terminal kinase (SAP/JNK) signaling pathway by phosphorylating different JNK isoforms. MKK7 is activated by upstream MKKKs, whereby the MKKKs interact with MKK7 through its C-terminal DVD domain (a.a. 377-400) and activate MKK7 by phosphorylating serine and threonine residues at a S-K-A-K-T motif located in the MKK7 kinase domain (a.a. 120-380). Upon activation, MKK4 and MKK7 in turn phosphorylate specific tyrosine and threonine residues of the conserved T-P-Y motif in the activation loop of substrate JNK proteins. The N-terminal D domain (a.a. 37-57), consisting of a cluster of positively charged amino acids followed by alternating hydrophobic amino acids, is required for the recruitment of MAPKK substrates, such as JNK. Alternative splicing results in four isoforms. Isoform B (beta), gamma, and isoform 4 differ from the canonical isoform A (alpha) by a 43-a.a. expansion at a.a. 111, a 16-a.a. expansion at a.a. 42, and a 7-a.a. expansion at a.a. 312, respectively, of the MKK7alpha sequence.
Specificity
Clone 3F6.5 was raised against recombinant N-terminal fragment derived from human MKK7/SKK4 spliced isoform 1 (isoform A or alpha) and isoform 4. The corresponding region in mouse isoform 2 & 8 (isoform alpha & beta 2) differ from the human sequence by only one amino acid (human A69 vs. mouse T69) Cross-reactivity toward other human and murine spliced isoforms is highly expected, but has not been confirmed.
